FDA approves 1st new tuberculosis drug in 40 years

(AP) ? The Food and Drug Administration on Monday approved a Johnson & Johnson tuberculosis drug that is the first new medicine to fight the deadly infection in more than four decades.

The agency approved J&J's pill, Sirturo, for use with older drugs to fight a hard-to-treat strain of tuberculosis that has not responded to other medications. However, the agency cautioned that the drug carries risks of potentially deadly heart problems and should be prescribed carefully by doctors.

Roughly one-third of the world's population is estimated to be infected with the bacteria causing tuberculosis. The disease is rare in the U.S., but kills about 1.4 million people a year worldwide. Of those, about 150,000 succumb to the increasingly common drug-resistant forms of the disease. About 60 percent of all cases are concentrated in China, India, Russia and Eastern Europe.

Sirturo, known chemically as bedaquiline, is the first medicine specifically designed for treating multidrug-resistant tuberculosis. That's a form of the disease that cannot be treated with at least two of the four primary antibiotics used for tuberculosis.

The standard drugs used to fight the disease were developed in the 1950s and 1960s.

"The antibiotics used to treat it have been around for at least 40 years and so the bacterium has become more and more resistant to what we have," said Chrispin Kambili, global medical affairs leader for J&J's Janssen division.

The drug carries a boxed warning indicating that it can interfere with the heart's electrical activity, potentially leading to fatal heart rhythms.

"Sirturo provides much-needed treatment for patients who have don't have other therapeutic options available," said Edward Cox, director of the FDA's antibacterial drugs office. "However, because the drug also carries some significant risks, doctors should make sure they use it appropriately and only in patients who don't have other treatment options."

Nine patients taking Sirturo died in company testing compared with two patients taking a placebo. Five of the deaths in the Sirturo group seemed to be related to tuberculosis, but no explanation was apparent for the remaining four.

Despite the deaths, the FDA approved the drug under its accelerated approval program, which allows the agency to clear innovative drugs based on promising preliminary results.

Last week, the consumer advocacy group Public Citizen criticized that approach, noting the drug's outstanding safety issues.

"The fact that bedaquiline is part of a new class of drug means that an increased level of scrutiny should be required for its approval," the group states. "But the FDA had not yet answered concerns related to unexplained increases in toxicity and death in patients getting the drug."

The FDA said it approved the drug based on two mid-stage studies enrolling 440 patients taking Sirturo. Both studies were designed to measure how long it takes patients to be free of tuberculosis.

Results from the first trial showed most patients taking Sirturo plus older drugs were cured after 83 days, compared with 125 days for those taking a placebo plus older drugs. The second study showed most Sirturo patients were cured after 57 days.

'The Hobbit' holds on to first place with $32M

(AP) ? A tiny hobbit has a mighty hold on the box office, staying on top for a third-straight week and wrapping up a year that saw a record-breaking $10.8 billion in total annual grosses.

The Warner Bros. fantasy "The Hobbit: An Unexpected Journey" remained at No. 1 with $32 million for a total of $221.7 million, according to final studio figures Monday. That's slightly down from the Sunday estimate of just under $33 million but enough to retain first place in a holiday weekend that featured the much-anticipated debuts of awards contenders "Django Unchained" and "Les Miserables."

The top 20 movies at U.S. and Canadian theaters Friday through Sunday, followed by distribution studio, gross, number of theater locations, average receipts per location, total gross and number of weeks in release, as compiled Monday by Hollywood.com are:

Cyber-bullying topic of Jan. 15 AAUW community forum

The annual Community Forum sponsored by the Coastal-Georgetown American Association of University Women will focus on cyber-bullying - what it is, what schools and organizations are doing to address the issue and what tools and resources are available to parents, teachers and children to assist them when confronting cyber-bullying.

Cosponsoring organizations include: Children & Families First, AAUW Delaware, Girls Inc. of DE, Girl Scouts of the Chesapeake Bay, Wilmington University, League of Women Voters of Sussex County and Delaware Bullying Prevention Association.

Cyber-bullying is a nationwide issue that affects all ages, but is particularly difficult for school-age children. National statistics show that about one-third of middle and high school students report being bullied and nearly one million school-age children are cyber-bullied annually

The AAUW Forum will be held at 7 p.m., Tuesday, Jan. 15, at Epworth UM Church, Holland Glade Road, Rehoboth.? No pre-registration is needed.

Delaware Attorney General Beau Biden will speak about current cyber-bullying legislation.

The featured speaker, Dr. Julius Mullen, chief clinical officer of Children & Families First, will speak about the increasing problem of cyber-bullying as it relates to awareness and prevention strategies from a parent and teacher perspective.? A panel of experts will address aspects of bullying. Panel members include John Sadowski, Delaware Department of Education; Coleen O?Connor, Delaware Bullying Prevention Association; and Chris Beagle, CAMP Rehoboth. Dr. Rebecca Ghabour, chair of psychology at Wilmington University, will moderate. Time will be set aside for questions and answers.

AAUW is committed to advancing equity for women and girls through advocacy, education, philanthropy and research.? The Coastal-Georgetown branch has a membership of over 90 women who actively pursue this mission through advocating at the state legislature for issues impacting women and children such as pay equity, affordable housing, foster care and early education.

4 dead in Moscow airliner crash

MOSCOW (AP) ? A passenger airliner careered off the runway at Russia's third-busiest airport and partly onto a highway while landing on Saturday, broke into pieces and caught fire, killing at least four people.

Officials said there were eight people aboard the Tu-204 belonging to Russian airline Red Wings that was flying back from the Czech Republic without passengers to its home at Vnukovo Airport.

Emergency officials said in a televised news conference that four people were killed and another four severely injured when the plane rolled off the runway into a snowy field and partly onto an adjacent highway, then disintegrated. No collisions with vehicles on the major, multilane highway were reported.

The plane's cockpit area was sheared off from the fuselage and a large chunk gashed out near the tail.

The crash occurred amid light snow and winds gusting up to 15 meters a second (30 mph), but other details were not immediately known. A spokesman for Russia's top investigative agency, Vladimir Markin, said initial indications were that pilot error was the cause.

The state news agency RIA Novosti cited an unidentified official at the Russian Aviation Agency as saying another Red Wings Tu-204 had gone off the runway at the international airport in Novosibirsk in Siberia on Dec. 20. The agency said that incident, in which no one was injured, was due to the failure of the plane's engines to go into reverse upon landing and that its brake system malfunctioned.

On Friday, the Aviation Agency sent a directive to the Tupolev company's president calling for it to take urgent preventive measures.

Vnukovo airport spokeswoman Yelena Krylova said it had enough personnel and equipment to keep the runway fully functional Saturday. The airport resumed receiving planes after a break of several hours.

Prior to Saturday's crash, there had been no fatal accidents reported for Tu-204s, which entered commercial service in 1995. The plane is a twin-engine midrange jet with a capacity of about 210 passengers.

Vnukovo, on the southern outskirts of Moscow, is one of the Russian capital's three international airports.

State Dept. warns Americans about Haiti travel

WASHINGTON (AP) ? The State Department has issued a revised Haiti travel advisory, warning Americans planning to travel to the Caribbean island nation about robbery, lawlessness, infectious disease and poor medical facilities.

"U.S. citizens have been victims of violent crime, including murder and kidnapping, predominantly in the Port-au-Prince area. No one is safe from kidnapping, regardless of occupation, nationality, race, gender or age," the department said.

The new travel warning was released Friday to replace a less strongly worded advisory issued in June.

In recent months, travelers arriving in Port-au-Prince, the capital and largest city, on flights from the United States have been attacked and robbed after leaving the airport. This year, at least two U.S. citizens were shot and killed in robbery and kidnapping incidents, the State Department said.

"Haitian authorities have limited capacity to deter or investigate such violent acts or prosecute perpetrators," the department said.

The State Department also noted that while the incidents of cholera have declined, the disease persists in many areas of Haiti. Medical facilities, including ambulance services, are particularly weak.

"Thousands of U.S. citizens safely visit Haiti each year, but the poor state of Haiti's emergency response network should be carefully considered when planning travel. Travelers to Haiti are encouraged to use organizations that have solid infrastructure, evacuation and medical support options in place," the department said.

I tested this game out on my own family over the Christmas holiday?and had three generations playing happily together. PORTLAND, Maine (AP) ? After waiting years and seeing marriage rights nearly awarded and then retracted, gay couples in Maine's largest city didn't have to wait a moment longer than necessary to wed, with licenses issued at the stroke of midnight as the law went into effect.

Among them were Steven Bridges and Michael Snell, who held a commitment ceremony six years ago but made marriage official under state law with a simple ceremony.

"It's historic. We've waited our entire lives for this," said Bridges, a retail manager, who's been in a relationship with the Snell, a massage therapist, for nine years. Bridges, 42, and Snell, 53, wore lavender and purple carnations on black T-shirts with the words "Love is love."

With Snell's two adult daughters looking on, they exchanged vows in the city clerk's office after getting the first marriage license issued to a same-sex couple in Portland.

Voters approved gay marriage in November, making Maine and two other states the first to do so by popular vote. The law is already in effect in Washington state; Maryland's takes effect on Tuesday, the first day of 2013.

Gay marriage was already legal in New York, Connecticut, Iowa, Massachusetts, New Hampshire, Vermont and the District of Columbia, but those laws were either enacted by lawmakers or through court rulings.

The Maine Legislature had once approved same-sex marriage, but it was overturned by a statewide referendum three years ago, crushing couples who had already made wedding plans. Gay marriage supporters collected signatures to put it on the ballot again, and this time it was easily approved.

Gov. Paul LePage signed off on the certified election results on Nov. 29, so the new law was to go into effect 30 days from that date. In addition to gay marriage becoming legal, same-sex marriages in other states will now be recognized by the state of Maine.

Nobody knew exactly how many couples would be rushing to get their marriage licenses early Saturday. Falmouth joined Portland in opening at midnight. A handful of other communities including Bangor, Brunswick and Augusta planned to hold special Saturday hours.

In Portland, the mood was festive with the crowd cheering and horns sounding at midnight as Bridges and Snell began filling out paperwork in the clerk's office in Portland City Hall.

More than a dozen couples stood in line to wait their turn amid the festive atmosphere. There were free carnation boutonnieres and cupcakes, and a jazz trio played.

Outside City Hall, a couple hundred people cheered Snell and Bridges when they emerged newly married, breaking into an impromptu song, the Beatles' "All You Need is Love."

Donna Galluzzo and Lisa Gorney also planned a midnight wedding, and theirs had all the trappings. They dined Friday night with friends, and then took a limo to City Hall. They had their rings, flowers, wedding vows and a friend to perform the ceremony.

They ended up near the back of the line awaiting marriage licenses, but that didn't matter.

"We decided it's a historic day and we thought it would be awesome to be a small part of history, to say we got married on the first day it's legal," Galluzzo said.

Not everyone was getting married right away.

Suzanne Blackburn and Joanie Kunian, of Portland, were among those in line to get their license at midnight, but they didn't plan to wed immediately. One of their grandchildren wanted them to get married on Valentine's Day.

"I don't think that we dared to dream too big until we had the governor's signature," Blackburn said. "That's why it's so important, because it feels real."

N.Y. woman charged in slaying of firefighters

WEBSTER, N.Y. (AP) ? A 24-year-old woman was arrested Friday and charged in connection with the Christmas Eve ambush slaying of two volunteer firefighters responding to a house fire in upstate New York.

Dawn Nguyen of Rochester faces a state charge of filing a falsified business record, State Police Senior Investigator James Sewell said. Nguyen also faces a federal charge; a news conference with the U.S. attorney in western New York was scheduled for 4 p.m. Friday.

Sewell said the state charge is connected to the purchase of an AR-15 semiautomatic rifle and a 12-gauge shotgun that William Spengler had with him Monday when firefighters Michael Chiapperini and Tomasz Kaczowka were gunned down. Three other people were wounded before the 62-year-old Spengler killed himself. He also had a .38-caliber revolver, but Nguyen is not connected to that gun, Sewell said.

The .223-caliber Bushmaster rifle, which had a combat-style flash suppressor, is similar to the one used by the gunman who massacred 20 children and six women in a Newtown, Conn., elementary school earlier this month.

Nguyen and her mother, Dawn Welsher, lived next door to Spengler in 2008. On Wednesday and again on Friday, shortly before her arrest, she answered her cellphone and told The Associated Press that she didn't want to talk about Spengler. Her brother, Steven Nguyen, told the Democrat and Chronicle newspaper of Rochester that Spengler stole the guns from Dawn Nguyen.

A number listed in the name of her lawyer, David Palmiere, was disconnected.

Spengler set a car on fire and touched off an inferno in his Webster home on a strip of land along the Lake Ontario shore, took up a sniper's position and opened fire on the first firefighters to arrive at about 5:30 a.m. on Christmas Eve, authorities said. He wounded two other firefighters and an off-duty police officer who was on his way to work.

A Webster police officer who had accompanied the firefighters shot back at Spengler with a rifle in a brief exchange of gunfire before the gunman killed himself.

Spengler spent 17 years in prison for killing his grandmother in 1980 and was barred from possessing weapons as a convicted felon.

Investigators still haven't released the identity of remains found in William Spengler's burned house. They have said they believe the remains are those of his 67-year-old sister, Cheryl Spengler, who also lived in the house near Rochester and has been unaccounted for since the killings. The Spengler siblings had lived in the home with their mother, Arline Spengler, who died in October. In all, seven houses were destroyed by the flames.

Investigators found a rambling, typed letter laying out Spengler's intention to destroy his neighborhood and "do what I like doing best, killing people."

He had been released from parole in 2006 on the manslaughter conviction, and authorities said they had had no encounters with him since.

I don't need a plastic window, or something that looks like it was designed by aliens -- just something that's sharp, solid, good looking and has a respectable airflow. Is that too much to ask?"

Well, we trawled our brains (as well as a fair chunk of the internet) looking for some classy chassis, and this is what we found:

  • Corsair's Obsidian 550D may have a plastic window, but otherwise it's a monolithic piece of imposing black metal that won't ugly up your office.
  • Coolermaster's Silencio 650 is hewn from diamond-cut aluminum and drops the plastic window, and given the company's heritage, should have excellent airflow.
  • Finally, there's the Lian-Li PC-A71F, which can only be described as terrifyingly sleek.

Record set in solar cell efficiency - with the light of a thousand suns

Scientists at the U.S. Department of Energy have created a solar cell that can convert 44 percent of sunlight hitting it into electrical energy, setting a new record for solar cell?efficiency. But it was only achieved by multiplying the power of the sun by nearly 1,000.

The ongoing research is being done at the III-V Multijunction Photovoltaics group at the Department of Energy's National Renewable Energy Laboratory, where the previous record of 43.5 percent was set. So while it's not a huge leap in efficiency, it's still a record.

The researchers achieve these high efficiencies by stacking multiple "junctions" into a single solar cell, each of which absorbs light at a slightly different wavelength. The "III-IV" in the name refers to the fact that the elements used?come from the third and fifth columns of the periodic table.

A new material with many desirable properties was?discovered by accident (gallium arsenide diluted with nitrogen), but proved extremely difficult to manufacture. Researchers needed to use a technique called molecular beam epitaxy to minimize impurities in the material ? but no one in the world could do it.

Solar Junction, a company started by researchers from Stanford University, decided to take up the challenge. With a few million dollars in funding, Solar Junction and NREL managed to make the new process work. The result was the "SJ3" cell, which has been breaking records ever since.

The catch is that it's not like a traditional solar cell, which simply receives sunlight on its surface and creates a modest charge from the photons. The SJ3 cell requires the power of the sun to be multiplied by special lenses, creating a beam of light hundreds of times brighter than ordinary sunlight.

In this kind of system, the power is measured in "suns": A beam 10?times brighter than the sun would be described simply as 10 suns. And?the previous efficiency record was set by a lens system that created 415 suns. The latest record was set using 947 suns.

Naturally, one may wonder if such a design is practical ? after all, how often does the sun shine a thousand times brighter than normal? But the lenses actually take normal sunlight that might have hit in a diffuse way on normal cells and concentrate it onto the more efficient SJ3 cell. So in the end it's just using the same area of sunlight in a different way.

The NREL emphasized in the report describing the record that the SJ3 design can easily replace solar systems used in satellites and other space-bound machines ? but it's lighter and more efficient, reducing rocket load and allowing for more onboard electronics.

What's next? Tweaking the design to include a germanium layer used previously could raise efficiency as high as 50 percent ? definitely?a major landmark in solar power, if researchers?can achieve it, although applying it to everyday power needs is another project?altogether.

Beijing ? China?s new communist leaders are increasing already tight controls on Internet use and electronic publishing following a spate of embarrassing online reports about official abuses.

The measures suggest China?s new leader, Xi Jinping, and others who took power in November share their predecessors? anxiety about the Internet?s potential to spread opposition to one-party rule and their insistence on controlling information despite promises of more economic reforms.

"They are still very paranoid about the potentially destabilizing effect of the Internet," said Willy Lam, a politics specialist at the Chinese University of Hong Kong. "They are on the point of losing a monopoly on information, but they still are very eager to control the dissemination of views."

This week, China?s legislature took up a measure to require Internet users to register their real names, a move that would curtail the Web?s status as a freewheeling forum to complain, often anonymously, about corruption and official abuses. The legislature scheduled a news conference Friday to discuss the measure, suggesting it was expected to be approved.

That comes amid reports Beijing might be disrupting use of software that allows Web surfers to see sites abroad that are blocked by its extensive Internet filters. At the same time, regulators have proposed rules that would bar foreign companies from distributing books, news, music and other material online in China.

Beijing promotes Internet use for business and education but bans material deemed subversive or obscene and blocks access to foreign websites run by human rights and Tibet activists and some news outlets. Controls were tightened after social media played a role in protests that brought down governments in Egypt and Tunisia.

In a reminder of the Web?s role as a political forum, a group of 70 prominent Chinese scholars and lawyers circulated an online petition this week appealing for free speech, independent courts and for the ruling party to encourage private enterprise.

Xi and others on the party?s ruling seven-member Standing Committee have tried to promote an image of themselves as men of the people who care about China?s poor majority. They have promised to press ahead with market-oriented reforms and to support entrepreneurs but have given no sign of support for political reform.

Communist leaders who see the Internet as a source of economic growth and better-paid jobs were slow to enforce the same level of control they impose on movies, books and other media, apparently for fear of hurting fledgling entertainment, shopping and other online businesses.

Until recently, Web surfers could post comments online or on microblog services without leaving their names.

That gave ordinary Chinese a unique opportunity to express themselves to a public audience in a society where newspapers, television and other media are state-controlled. The most popular microblog services say they have more than 300 million users and some users have millions of followers reading their comments.

The Internet also has given the public an unusual opportunity to publicize accusations of official misconduct.

A local party official in China?s southwest was fired in November after scenes from a videotape of him having sex with a young woman spread quickly on the Internet. Screenshots were uploaded by a former journalist in Beijing, Zhu Ruifeng, to his Hong Kong website, an online clearing house for corruption allegations.

Some industry analysts suggest allowing Web surfers in a controlled setting to vent helps communist leaders stay abreast of public sentiment in their fast-changing society. Still, microblog services and online bulletin boards are required to employ censors to enforce content restrictions. Researchers say they delete millions of postings a day.

The government says the latest Internet regulation before the National People?s Congress is aimed at protecting Web surfers? personal information and cracking down on abuses such as junk e-mail. It would require users to report their real names to Internet service and telecom providers.

The main ruling party newspaper, People?s Daily, has called in recent weeks for tighter Internet controls, saying rumors spread online have harmed the public. In one case, it said stories about a chemical plant explosion resulted in the deaths of four people in a car accident as they fled the area.

Proposed rules released this month by the General Administration of Press and Publications would bar Chinese-foreign joint ventures from publishing books, music, movies and other material online in China. Publishers would be required to locate their servers in China and have a Chinese citizen as their local legal representative.
